The air that's taken out needs to be replaced somehow. If it's not by the same amount, the extraction rate will suffer. In industrial situations, this is fine by a lossnay heat exchanger, the air going out heats the air coming in. It would be hard to hear exchange in such a small space, so either have a supply duct that you can open it close or crack open a window to replace the air that is being sucked out.

No need for an air supply, although putting a vent in would be good anyway to allow the structure to breath properly anyway. I dont use internal fans to direct air to the extractor fan myself, a sufficiently sized fan should move enough air by itself, 4" bathroom fans are not powerful enough however.
I find my summerhouse holds its heat really well, a little electric fire is perfect. I insulated mine fully, but roof and floor minimum if you want it to be comfortable all year round.
